In his first keynote address on the issue in a few weeks, he said the US intention was to use the conflict as a reason to impose more sanctions on Russia.
He also said that the US was ignoring Russia's concerns about NATO allies in Europe.
The situation is tense due to the construction of Russian troops near the Ukrainian border.
Russia in recent weeks has deployed about 100,000 troops - fully armed from tanks and artillery to ammunition and air force - to the Ukrainian border.
But Russia denies Western allegations that it plans to invade, almost eight years after it took control of the Crimean peninsula in southern Ukraine and backed a bloody uprising in the eastern region of Donbas.
Moscow, meanwhile, blames the Ukrainian government for failing to ratify an international peace agreement in the east, in which at least 14,000 people have been killed and Russian-backed rebels control much of the country.
Meanwhile, Ukrainian President Volodymyr Zelensky warned on Tuesday that the Russian offensive "would not be a war between Ukraine and Russia - this would be a European war, a full-blown war.
Speaking after talks with Hungarian Prime Minister Viktor Orban in Moscow, Mr Putin said: "It seems to me that the United States is not so concerned about the security of Ukraine ... but its main task is to embrace Russia's development. consider Ukraine itself a mere tool for achieving this goal. "
The conflict between Russia and the US, which now has the largest nuclear arsenals in the world, dates back to the Cold War (1947-89). At that time Ukraine was an important part of the Communist Soviet Union, following Russia.
Mr Putin said the US was ignoring Moscow's concerns in responding to Russia's demands for legally binding security guarantees, including a ban on Nato expansion in the east.
He suggested that if Ukraine could not comply with its desire to join Nato, it could attract more members to the war with Russia.
"Imagine that Ukraine is a member of Nato and that the [Crimean] recapture of the Crimean army begins," the Russian leader said. "What - are we going to fight Nato? Has anyone ever thought about this? It seems they never did."
In Ukraine itself, UK Prime Minister Boris Johnson accused Mr Putin of "holding a gun ... in the head of Ukraine" and called on the Kremlin to back off from "military disaster".
Speaking after talks with Mr Zelensky in the capital Kyiv, Mr Johnson told reporters that Ukrainian troops would fight in the event of an attack.
"There are 200,000 men and women under arms in Ukraine," he said. "They are going to be very, very aggressive and bloody and I think parents, mothers, in Russia, should think about that fact. And I very much hope that President Putin backs away from the conflict and that we have engaged in dialogue."
Mr Johnson warned that the UK would respond to Russian violence "by imposing sanctions and alternatives when Russia's first toecap crosses into Ukraine".
The UK has announced it is donating £ 88m ($ 119) to Ukraine to promote Russia's stable governance and independence.
Mr Zelensky called for sanctions to be imposed before the increase, saying he would support any UK move to deal with "money laundering" allegedly linked to the Kremlin in London.
